C\u00e1c d\u00f2ng n\u00e0y c\u00f3 sinh tr\u01b0\u1edfng nhanh r\u00f5 r\u1ec7t h\u01a1n so v\u1edbi gi\u1ed1ng \u0111\u1ed1i ch\u1ee9ng U<sub>sx<\/sub> (\u0111\u1ed9 v\u01b0\u1ee3t tr\u1ed9i so v\u1edbi gi\u1ed1ng \u0111\u1ea1i tr\u00e0 v\u1edbi chi\u1ec1u cao &gt; 25% v\u00e0 \u0111\u01b0\u1eddng k\u00ednh &gt; 10%).\u00a0 Trong \u0111\u00f3 d\u00f2ng UU89 c\u00f3 kh\u1ea3 n\u0103ng t\u00e1i sinh cao n\u00ean \u0111\u01b0\u1ee3c s\u1eed d\u1ee5ng l\u00e0m v\u1eadt li\u1ec7u bi\u1ebfn n\u1ea1p gen.<\/p>\n<ul>\n<li>Ph\u00e2n l\u1eadp gen v\u00e0 x\u00e2y d\u1ef1ng c\u1ea5u tr\u00fac vector mang gen m\u1ee5c ti\u00eau<\/li>\n<\/ul>\n<p>&#8211;\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 Gen t\u0103ng chi\u1ec1u d\u00e0i s\u1ee3i g\u1ed7 (<i>EcHB1<\/i>) \u0111\u01b0\u1ee3c ph\u00e2n l\u1eadp t\u1eeb B\u1ea1ch \u0111\u00e0n camal (<i>Eucalyptus camaldulensis<\/i>). Tr\u00ecnh t\u1ef1 gen <i>EcHB1<\/i> c\u00f3 m\u1ee9c \u0111\u1ed9 t\u01b0\u01a1ng \u0111\u1ed3ng 100% so v\u1edbi tr\u00ecnh t\u1ef1 nucleotide c\u1ee7a gen <i>EcHB1<\/i> tr\u00ean Ng\u00e2n h\u00e0ng gen Qu\u1ed1c t\u1ebf.<\/p>\n<p>&#8211;\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 Gen <i>EcHB1<\/i> \u0111\u01b0\u1ee3c g\u1eafn v\u1edbi vector nh\u1ecb th\u1ec3 pGWB2, \u0111\u1ec3 t\u1ea1o th\u00e0nh c\u1ea5u tr\u00fac pGWB2-EcHB1. Vector pGWB2 mang gen ch\u1ecdn l\u1ecdc kh\u00e1ng sinh Kanamycine v\u00e0 Hygromycin<\/p>\n<p>&#8211;\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 Ho\u1ea1t \u0111\u1ed9ng c\u1ee7a gen <i>EcHB1<\/i> \u0111\u01b0\u1ee3c \u0111i\u1ec1u khi\u1ec3n b\u1edfi promoter CaMV 35S.<\/p>\n<p>&#8211;\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 C\u00e1c c\u1ea5u tr\u00fac vector chuy\u1ec3n gen pGWB2-EcHB1 \u0111\u01b0\u1ee3c bi\u1ebfn n\u1ea1p v\u00e0o vi khu\u1ea9n<i> Agrobacterium tumefaciens<\/i> ch\u1ee7ng C58-GV3101.<\/p>\n<ul>\n<li>\u0110\u00e3 ki\u1ec3m tra m\u1ee9c \u0111\u1ed9 bi\u1ec3u hi\u1ec7n c\u1ee7a gen m\u1ee5c ti\u00eau <i>EcHB1<\/i> v\u1edbi c\u1ea5u tr\u00fac vector \u0111\u01b0\u1ee3c x\u00e2y d\u1ef1ng tr\u00ean c\u00e2y Thu\u1ed1c l\u00e1 chuy\u1ec3n gen sau 3 th\u00e1ng tr\u1ed3ng t\u1ea1i v\u01b0\u1eddn \u01b0\u01a1m. K\u1ebft qu\u1ea3 cho th\u1ea5y sinh tr\u01b0\u1edfng c\u00e2y Thu\u1ed1c l\u00e1 chuy\u1ec3n gen h\u01a1n g\u1ea5p 1,8 l\u1ea7n c\u00e2y \u0111\u1ed1i ch\u1ee9ng v\u00e0 s\u1ee3i g\u1ed7 d\u00e0i h\u01a1n 1,2 l\u1ea7n so v\u1edbi c\u00e2y kh\u00f4ng chuy\u1ec3n gen.<\/li>\n<\/ul>\n<p><b>1.2.\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 <\/b><b>X\u00e2y d\u1ef1ng c\u00e1c quy tr\u00ecnh k\u1ef9 thu\u1eadt ph\u1ee5c v\u1ee5 c\u00f4ng t\u00e1c bi\u1ebfn n\u1ea1p gen<\/b><\/p>\n<ul>\n<li>\u00d8 \u0110\u00e3 x\u00e2y d\u1ef1ng quy tr\u00ecnh k\u1ef9 thu\u1eadt t\u00e1i sinh d\u00f2ng B\u1ea1ch \u0111\u00e0n lai UU89 v\u1edbi hi\u1ec7u su\u1ea5t t\u00e1i sinh cao th\u00f4ng qua ph\u00f4i soma (53,2% cho \u0111o\u1ea1n th\u00e2n v\u00e0 62,9% cho m\u1ea3nh l\u00e1) v\u00e0 th\u00f4ng qua \u0111a ch\u1ed3i (75,5% cho m\u1ea3nh l\u00e1 v\u00e0 71,8% cho \u0111o\u1ea1n th\u00e2n.<\/li>\n<li>\u00d8 \u0110\u00e3 x\u00e2y d\u1ef1ng \u0111\u01b0\u1ee3c quy tr\u00ecnh k\u1ef9 thu\u1eadt chuy\u1ec3n gen <i>EcHB1<\/i> cho d\u00f2ng B\u1ea1ch \u0111\u00e0n lai UU89 v\u1edbi hi\u1ec7u su\u1ea5t chuy\u1ec3n gen l\u00e0 1,06%.<\/li>\n<\/ul>\n<p><b>1.3.\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 <\/b><b>\u00a0Chuy\u1ec3n gen m\u1ee5c ti\u00eau <i>EcHB1<\/i> v\u00e0o B\u1ea1ch \u0111\u00e0n lai UU<\/b><\/p>\n<ul>\n<li>\u0110\u00e3 t\u1ea1o \u0111\u01b0\u1ee3c 01 d\u00f2ng B\u1ea1ch \u0111\u00e0n lai mang gen <i>EcHB1<\/i> sinh tr\u01b0\u1edfng b\u00ecnh th\u01b0\u1eddng trong \u0111i\u1ec1u ki\u1ec7n v\u01b0\u1eddn \u01b0\u01a1m v\u00e0 \u0111\u01b0\u1ee3c x\u00e1c \u0111\u1ecbnh mang gen <i>EcHB1<\/i> b\u1eb1ng ph\u01b0\u01a1ng ph\u00e1p PCR, lai Southern Blot.<\/li>\n<li>\u00d8 \u0110\u00e3 ph\u00e2n t\u00edch m\u1ee9c \u0111\u1ed9 bi\u1ec3u hi\u1ec7n c\u1ee7a gen <i>EcHB1<\/i> trong d\u00f2ng c\u00e2y chuy\u1ec3n gen b\u1eb1ng ph\u01b0\u01a1ng ph\u00e1p RT-PCR v\u00e0 Real Time qRT-PCR. K\u1ebft qu\u1ea3 ph\u00e2n t\u00edch cho th\u1ea5y d\u00f2ng E1 chuy\u1ec3n gen c\u00f3 m\u1ee9c \u0111\u1ed9 bi\u1ec3u hi\u1ec7n gen cao g\u1ea5p 100 l\u1ea7n so v\u1edbi d\u00f2ng \u0111\u1ed1i ch\u1ee9ng kh\u00f4ng chuy\u1ec3n gen.<\/li>\n<\/ul>\n<p><b>1.4.\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 <\/b><b>\u00a0\u0110\u00e1nh gi\u00e1 c\u00e2y B\u1ea1ch \u0111\u00e0n chuy\u1ec3n gen \u1edf giai \u0111o\u1ea1n v\u01b0\u1eddn \u01b0\u01a1m.<\/b><\/p>\n<ul>\n<li>D\u00f2ng B\u1ea1ch \u0111\u00e0n chuy\u1ec3n gen sau 5 th\u00e1ng tr\u1ed3ng t\u1ea1i v\u01b0\u1eddn \u01b0\u01a1m c\u00f3 sinh tr\u01b0\u1edfng nhanh h\u01a1n c\u00e2y \u0111\u1ed1i ch\u1ee9ng.<\/li>\n<li>Ph\u00e2n t\u00edch m\u1ed9t s\u1ed1 ch\u1ec9 ti\u00eau v\u1ec1 gi\u1ea3i ph\u1eabu g\u1ed7 t\u1ea1i giai \u0111o\u1ea1n 5 th\u00e1ng cho th\u1ea5y d\u00f2ng B\u1ea1ch \u0111\u00e0n chuy\u1ec3n gen E1 c\u00f3 s\u1ed1 l\u01b0\u1ee3ng m\u1ea1ch g\u1ed7, tia g\u1ed7 v\u00e0 s\u1ee3i g\u1ed7 d\u00e0i h\u01a1n c\u00e2y \u0111\u1ed1i ch\u1ee9ng.
